目的:研究核转录因子-κB(nuclear factor-κB,NF-κB)和环氧化酶-2(cyclooxygenase-2,COX-2)在舌鳞状细胞癌(tongue squamous cell carcinoma,TSCC)发生、发展中的作用及两者间的关系。方法:取经手术病理证实的舌鳞状细胞癌标本50例,采用SP免疫组化法检测舌鳞状细胞癌组织中NF-κB和COX-2的表达情况。应用SPSS11.0软件包,采用χ2检验分析NF-κB和COX-2表达的关系及其与临床病理特征的关系。结果:低分化TSCC组织中NF-κB的阳性表达率为48%,显著高于中、高分化的癌组织(P<0.05);有淋巴结转移的TSCC组织中,NF-κB的阳性表达率为42.3%,显著高于无淋巴结转移的患者(P<0.05)。低分化TSCC组织中,COX-2的阳性表达率为68.0%,显著高于中、高分化者(P<0.05);有淋巴结转移组,COX-2的阳性表达率为61.5%,高于无淋巴结转移组(P<0.05)。在COX-2阴性表达的患者中,NF-κB阳性表达率为3.8%(1/26),显著低于COX-2阳性表达的患者中NF-κB阳性表达的比率(P<0.05)。结论:NF-κB和COX-2在低分化及有淋巴结转移的舌鳞状细胞癌中的阳性表达率均高于中、高分化及无淋巴结转移的病例,且两者间的表达具有相关性,NF-κB和COX-2可能参与了舌鳞状细胞癌的发生与发展。
Objective: To investigate the expression of nuclear factor-κB (NF-κB) and cyclooxygenase-2 (COX-2) in tongue squamous cell carcinoma (TSCC) The role of development and the relationship between the two. Methods: Totally 50 specimens of tongue squamous cell carcinoma confirmed by surgery and pathology were used to detect the expression of NF-κB and COX-2 in tongue squamous cell carcinoma by SP immunohistochemical method. The relationship between NF-κB and COX-2 expression and clinicopathological features were analyzed by χ2 test using SPSS11.0 software package. Results: The positive expression rate of NF-κB in poorly differentiated TSCC was 48%, which was significantly higher than that in moderately differentiated carcinoma (P <0.05). The positive rate of NF-κB in TSCC with lymph node metastasis was 42.3%, significantly higher than those without lymph node metastasis (P <0.05). In poorly differentiated TSCC tissues, the positive rate of COX-2 expression was 68.0%, which was significantly higher than that in moderately and moderately differentiated tissues (P <0.05). The positive rate of COX-2 in lymph node metastasis group was 61.5% Lymph node metastasis group (P <0.05). In COX-2-negative patients, the positive rate of NF-κB was 3.8% (1/26), which was significantly lower than that of COX-2 positive patients (P <0.05). CONCLUSION: The positive rates of NF-κB and COX-2 in poorly differentiated and squamous cell carcinoma with lymph node metastasis are higher than those in moderately and well differentiated and without lymph node metastasis, and the correlation between the expression of NF-κB and COX-2 , NF-κB and COX-2 may participate in the occurrence and development of tongue squamous cell carcinoma.
