目的探讨脱氧雪腐镰刀菌烯醇(DON)对胃癌细胞SGC-7901、BGC-823凋亡的影响及其可能机制。方法体外培养的胃癌细胞株SGC-7901、BGC-823细胞经100、500、1000μg/LDON处理72h,以流式细胞术(FCM)检测细胞凋亡情况及其量效关系,流式细胞术、免疫细胞化学和Westernblot检测凋亡相关蛋白Bax、Bcl-2蛋白的表达情况。结果FCM定量检测结果表明,DON各处理组细胞凋亡率均高于对照组,且均随DON浓度升高凋亡率相应增加,具有明显量效关系(SGC-7901:r=0.660,P<0.05,n=4;BGC-823:r=0.750,P<0.01,n=4)。FCM、免疫细胞化学和Westernblot结果显示,DON处理后SGC-7901、BGC-823细胞Bax蛋白表达上调,Bcl-2蛋白表达下降。结论DON可剂量依赖地诱导体外培养的胃癌细胞SGC-7901、BGC-823凋亡,其机制可能与上调Bax蛋白的表达,下调Bcl-2蛋白的表达有关。
Objective To investigate the effect of deoxynivalenol (DON) on the apoptosis of gastric cancer cells SGC-7901 and BGC-823 and its possible mechanism. Methods The gastric cancer cell lines SGC-7901 and BGC-823 were treated with 100, 500, 1000μg / LDon for 72 hours. Flow cytometry (FCM) was used to detect apoptosis and its dose-response relationship. Immunocytochemistry and Western blot were used to detect the expression of Bax and Bcl-2. Results FCM results showed that the apoptotic rate of all the treated groups was higher than that of the control group, and the apoptotic rate increased with the increase of DON concentration, with a significant dose-response relationship (SGC-7901: r = 0.660, P < 0.05, n = 4; BGC-823: r = 0.750, P <0.01, n = 4). FCM, immunocytochemistry and Western blot showed that Bax protein expression was up-regulated and Bcl-2 protein expression was down-regulated in DON-treated SGC-7901 and BGC-823 cells. Conclusion DON can dose-dependently induce the apoptosis of SGC-7901 and BGC-823 cells in vitro. The mechanism may be related to the up-regulation of Bax protein expression and the down-regulation of Bcl-2 protein expression.
