目的:分析十二指肠钩虫成虫及第三期幼虫(L3)的可溶性抗原的蛋白组分,并鉴定后者的特异性组分,为钩虫病免疫学的后继研究提供资料.方法:十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)及酶联免疫印迹技术(ELIB).结果与结论:十二指肠钩虫成虫可溶性抗原有21条蛋白带,主带的分子量分别是94、66、48、28、24、20、16、12和10kD,其中16和12kD两带着色尤为浓重;L3有22条蛋白带,主带分子量分别是94、63、60、52、50、47、41、39、32和17kD,钩虫病人血清可识别L3的5-12条区带,其中47和52kD两组分能被所有钩虫病人血清识别,而不与正常血清反应,初步说明它们具有特异性,提示有进一步研究的价值.“,”To analyze the protein components of the antigens of A. duodenale adult and L3 worms. METHOD:SDS-PAGE and ELIB(or immuno-blot text)were applied. RESULTS and CONCLUSION: Analysis by applying SDS-PAGE showed that the antigen of A. duodenale adult worm appeared 21 protein bands. The MW of the main bands were 94, 66, 48, 28, 24, 20, 16, 12 and 10kD respectively. It was noticeable that the antigen possess abundant low molecular mass polypepties ranging between 12~16kD molecular mass. The antigen of L3 worms appeared in 22 protein bands and the MW of the main bands were 94, 63, 60, 52, 50, 47, 41, 39, 32 and 17kD respectively. By using ELIB, 5~12 protein bands of L3 antigen ranging between 9~131kD molecular mass could be recognized separately by sera from 11 patients with hookworm infection,among which two bands with MW of 47kD and 52kD reacted to all positive sera,but not to the negative sera,inferring that they were of specificity.
