目的研究乌梅散对染镉所致小鼠肝脏和肾脏损伤的保护作用。方法昆明小鼠随机分为四组,每组10只,空白对照组、镉染毒组、乌梅散高剂量组和低剂量组。低剂量组灌胃给予乌梅散50mg/(kg·d),高剂量组100mg/(kg·d),各组每天灌胃一次,连续30d。镉染毒组和乌梅散组腹腔注射氯化镉3mg/(kg·d)染毒,连续染毒10d。第三十天,测定小鼠血清天冬氨酸转氨酶(AST)和丙氨酸转氨酶(ALT)活力,测定血清、肝肾组织中的超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量。观察肝组织和肾组织病理学变化。结果与空白组相比,镉染毒组小鼠血清中AST、ALT水平明显上升(P<0.05),血清、肝肾组织中SOD活力显著减低(P<0.05),而MDA含量明显升高(P<0.05)。与镉染毒组相比,乌梅散低、高剂量组小鼠血清中AST、ALT活力明显降低(P<0.05),血清、肝肾组织中SOD活性显著升高(P<0.05),MDA含量明显降低(P<0.05)。病理结果显示,镉染毒组小鼠肝细胞肿胀,有片状细胞环,肾近曲小管可见管腔形态不完整;乌梅散高剂量组肝细胞损伤程度有所减轻。乌梅散高剂量组小鼠近曲小管受损程度减轻。结论乌梅散对镉染毒所致的小鼠肝肾损伤有保护作用。
Objective To study the protective effect of WuMeiSan on the liver and kidney damage induced by cadmium in mice. Methods Kunming mice were randomly divided into four groups with 10 rats in each group, blank control group, cadmium poisoning group, high-dose wu mei powder group and low-dose group. The low-dose group was given intragastric administration of Wumeisan 50mg / (kg · d), high-dose group 100mg / (kg · d), gavage once daily for 30 days. Cadmium poisoning group and Wumeisan group were intraperitoneal injection of cadmium chloride 3mg / (kg · d) exposure, continuous exposure 10d. On the thirtieth day, the activities of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in serum of mice were measured. The activities of superoxide dismutase (SOD) and malondialdehyde (MDA) in serum, )content. Pathological changes of liver and kidney were observed. Results Compared with the blank control group, the levels of AST and ALT in the serum of cadmium-exposed mice increased significantly (P <0.05) and the activity of SOD in serum, liver and kidney decreased significantly (P <0.05) and MDA content increased significantly P <0.05). Compared with cadmium exposure group, the activities of AST and ALT in the serum of the mice in the low and high doses of Wumei San significantly decreased (P <0.05), while the activities of SOD in the serum, liver and kidney were significantly increased (P <0.05) Significantly lower (P <0.05). The pathological results showed that the liver cells in cadmium-exposed mice swollen with a lamellar ring, and the proximal tubule of the kidney showed incomplete lumen morphology. The injury of liver cells in the high-dose Wumensan group was lessened. Wumei powder high dose group of mice near the lesser damage to the tubule. Conclusion Wumeisan has a protective effect on liver and kidney injury induced by cadmium in mice.
