佛手酚、佛手柑内酯和花椒毒素为明党参中天然呋喃香豆素成分,具多重药理作用。以明党参栽培品和悬浮培养细胞为材料,研究茉莉酸甲酯处理后呋喃香豆素积累状况和一系列生理生化指标变化。结果显示,茉莉酸甲酯不同程度地促进明党参中各呋喃香豆素合成,柄悬浮细胞中产量高于叶悬浮细胞。于培养周期第10天加入100μmol·L-1茉莉酸甲酯对明党参悬浮细胞作用效果最明显,最适收获时间为处理4 d后,此时柄悬浮细胞中佛手酚,花椒毒素和佛手柑内酯产量分别是未诱导细胞的73.6,1.9,42.7倍。茉莉酸甲酯诱导呋喃香豆素增长的同时抑制细胞活力和生物量积累,激发细胞防御反应。该研究初次系统地探索了茉莉酸甲酯对明党参中呋喃香豆素的诱导效果,并为大量生产3种活性成分和探明茉莉酸甲酯诱导机制奠定基础。
Bergamot, bergamot and pricklyashin are the natural furanocoumarin components of Codonopsis Pilosula which have multiple pharmacological effects. Taking Codonopsis pilosula cultivated and suspension cultured cells as materials, the accumulation status of furocoumarin and the changes of a series of physiological and biochemical indexes after methyl jasmonate treatment were studied. The results showed that methyl jasmonate promoted the synthesis of furocoumarin in Codonopsis pilosula to varying degrees, and the yield of stem suspension cells was higher than that of leaf suspension cells. The effect of adding 100μmol·L-1 jasmonic acid methyl ester on the suspension cells of Cynanum cumingii at the 10th day of the culture cycle was the most obvious. The optimum harvesting time was 4 days after the treatment, and the contents of bergol, xanthoxins and bergamot in suspension cells The lactone yields were 73.6, 1.9, 42.7 times that of uninduced cells, respectively. Methyl jasmonate induces the growth of furocoumarins and inhibits cell viability and biomass accumulation, stimulating cellular defensive responses. This study, for the first time, systematically explored the induction effect of methyl jasmonate on furocoumarin in Codonopsis pilosula and laid the foundation for the mass production of three active ingredients and the mechanism of induction of methyl jasmonate.
