目的:研究酶解与溶剂联合提取洋葱中槲皮素的最佳工艺条件。方法:以槲皮素得率为考察指标,通过单因素试验及正交实验确定最佳提取工艺。结果:酶解-溶剂联合提取槲皮素的最佳工艺条件是:酶解pH为5.5,酶解浓度为0.5mg.ml-1,酶解温度为45℃,酶解时间为2 h,液固比为20∶1,提取温度为65℃,提取时间为1.5 h,乙醇浓度为60%,槲皮素平均得率为0.046%。结论:采用酶解-溶剂联合提取槲皮素,工艺方法简单,可大大提高原料利用率。
Objective: To study the optimum conditions for extracting quercetin from onion with enzymolysis and solvent. Methods: Taking quercetin yield as index, the best extraction technology was determined by single factor experiment and orthogonal experiment. Results: The optimal conditions for the extraction of quercetin were enzymolysis pH 5.5, enzymolysis concentration 0.5 mg.ml-1, enzymolysis temperature 45 ℃, enzymolysis time 2 h, The solid ratio was 20:1, the extraction temperature was 65 ℃, the extraction time was 1.5 h, the ethanol concentration was 60% and the average yield of quercetin was 0.046%. Conclusion: Enzymolysis - solvent extraction of quercetin, the process is simple, can greatly improve the utilization of raw materials.
