目的:对不同产地秦艽的质量和遗传多样性进行研究。方法:使用性状、显微鉴别方法进行鉴别研究,采用高效液相色谱法进行龙胆苦苷含量测定,利用RAPD技术对其遗传多样性和遗传结构进行研究。结果:表明不同产地秦艽的性状、显微特征具有差异,其有效成分龙胆苦苷的含量也具有一定的差异,各种群的遗传多样性水平依次为湟中种群(K)>环县种群(J)>子午岭种群(H)。结论:对不同地区秦艽药材的外部形态、内部构造、有效成分含量和遗传水平的顺序,为保护野生品种、确保引种栽培秦艽药材的质量提供了科学依据。
Objective: To study the quality and genetic diversity of Gentiana macrophylla from different areas. Methods: The characters of the gentiopicroside were determined by the method of microscopic identification. The content of gentiopicroside was determined by high performance liquid chromatography. The genetic diversity and genetic structure of gentiopicroside were studied by RAPD technique. Results: The results showed that the traits and microscopic features of Gentiana macrophylla in different areas were different, and the content of Gentiopicroside in Gentiana macrophylla was also different. The genetic diversity of each genus was in order of population (K)> Huanxian population (J)> Ziwuling population (H). Conclusion: The sequence of external morphology, internal structure, active ingredients and genetic level of Gentiana macrophylla in different regions provides a scientific basis for protecting wild species and ensuring the quality of Gentiana macrophylla.
