目的:建立藏紫菀提取物中总黄酮及槲皮素、木犀草素的含量测定方法。方法:以芦丁为对照品,采用紫外分光光度法,以亚硝酸钠-硝酸铝显色,在510 nm处测定藏紫菀提取物中总黄酮的含量。采用高效液相色谱法同时测定藏紫菀提取物中槲皮素、木犀草素的含量。色谱柱为Waters symmetry C18柱,流动相为甲醇-0.1%磷酸溶液(55∶45),检测波长为360 nm,体积流量1.0 m L/min,柱温30℃,进样量10μL。结果:芦丁在浓度4.12μg/m L~41.2μg/m L的范围内与吸光度值呈良好的线性关系(r=0.9996),平均加样回收率为98.37%,(RSD%=2.04%,n=9)。槲皮素在0.0736~0.736μg(r=0.9993),木犀草素在0.0444~0.444μg(r=0.9994)的范围内呈良好的线性关系。平均回收率槲皮素为99.31%((RSD%=1.72%,n=9),木犀草素为98.90%(RSD%=1.81%,n=9)。结论:本法操作简便,快速准确,可作为藏紫菀提取物中黄酮类物质的含量测定方法。
Objective: To establish a method for the determination of total flavonoids, quercetin and luteolin in Tibetan aster. Methods: Rutin was used as reference substance, and the content of total flavonoids in Aster euphorbiae rubra was determined by UV spectrophotometry with sodium nitrite-aluminum nitrate colorimetry at 510 nm. Simultaneous determination of quercetin and luteolin in Tibetan kryptonum by high performance liquid chromatography. The chromatographic column was a Waters symmetry C18 column with a mobile phase of methanol-0.1% phosphoric acid (55:45) at a detection wavelength of 360 nm with a volume flow rate of 1.0 mL / min and a column temperature of 30 ° C. RESULTS: Rutin had a good linear relationship with absorbance (r = 0.9996) in the concentration range of 4.12μg / m L to 41.2μg / m L, and the average recovery was 98.37% (RSD% = 2.04% n = 9). Quercetin had a good linear relationship in the range of 0.0736 ~ 0.736μg (r = 0.9993) and luteolin in the range of 0.0444 ~ 0.444μg (r = 0.9994). The average recoveries of quercetin were 99.31% (RSD% = 1.72%, n = 9) and luteolin was 98.90% (RSD% = 1.81%, n = 9) .Conclusion: The method is simple, rapid and accurate, As a method for the determination of the content of flavonoids in Tibetan kaffir extract.
