目的 CD4+CD25+Foxp3+调节性T细胞(CD4+CD25+Foxp3+regulatory Tcells,Treg)在自身免疫耐受、移植和抗肿瘤应答中起着重要的作用,它们在肿瘤患者中的作用机制尚未阐明。本研究分析、探讨肝癌患者肿瘤组织和外周血CD4+CD25+Foxp3+调节性T细胞频率的临床意义及其生存率的关系分析。方法用流式细胞仪分析肝癌患者的肿瘤组织、外周血中单个核细胞(PBMCs)的CD4+CD25+Foxp3+调节性T细胞频率,并回顾分析20例肝癌患者,均随访3年,比较不同的CD4+CD25+Foxp3+调节性T细胞频率与生存率的关系。结果肝癌患者外周血和肝癌组织CD4+CD25+Foxp3+T细胞占PBMCs的比例均较对照组显著增高。并且CD4+CD25+Foxp3+T细胞的阳性率与肝癌的病理分期存在相关性。病理分期越晚,CD4+CD25+Foxp3+T细胞的阳性率越高,且与生存率负相关。结论 CD4+CD25+Foxp3+调节性T细胞在肝癌患者中比例升高,可能是肝癌患者细胞免疫功能削弱的机制之一。
Objective CD4 + CD25 + Foxp3 + regulatory T cells (Tregs) play an important role in autoimmune tolerance, transplantation and anti-tumor response, and their mechanism of action in tumor patients has not been elucidated . This study analyzed the relationship between the clinical significance and the survival rate of CD4 + CD25 + Foxp3 + regulatory T cells in tumor tissues and peripheral blood of patients with hepatocellular carcinoma. Methods The frequencies of CD4 + CD25 + Foxp3 + regulatory T cells in tumor tissues and peripheral blood mononuclear cells (PBMCs) of patients with hepatocellular carcinoma were analyzed by flow cytometry, and 20 patients with liver cancer were retrospectively analyzed. All patients were followed up for 3 years, Relationship between CD4 + CD25 + Foxp3 + regulatory T cells and survival rate. Results The proportion of CD4 + CD25 + Foxp3 + T cells in PBMCs in peripheral blood and hepatocellular carcinoma patients were significantly higher than those in control group. And the positive rate of CD4 + CD25 + Foxp3 + T cells was correlated with the pathological stage of HCC. The later the pathological stage, the higher the positive rate of CD4 + CD25 + Foxp3 + T cells, and negatively correlated with the survival rate. Conclusion The proportion of CD4 + CD25 + Foxp3 + regulatory T cells in HCC patients may be one of the mechanisms of impaired cellular immune function in HCC patients.
