目的观察苯中毒再生障碍性贫血(BPAA)患者血清中造血负调控因子对正常骨髓造血细胞的影响。方法以BPAA患者为研究对象,用其血清培养健康者骨髓细胞,进行骨髓造血干/祖细胞集落形成单位(CFU)计数,流式细胞仪检测CD34+细胞率及其Fas表达率的变化,同时用酶联免疫吸附法(ELISA)检测血清中干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)、白细胞介素-2(IL-2)的水平。结果 BPAA患者血清培养后骨髓粒-巨噬细胞集落形成单位(CFU-GM)和红系集落形成单位(CFU-E)数量均显著降低(均P<0.05),骨髓CD34+细胞比例显著降低,CD34+细胞的Fas表达率随之显著升高(均P<0.05);血清中IFN-γ、TNF-α、IL-2的水平也明显升高(均P<0.05)。结论 BPAA患者血清中造血负调控因子水平升高,使正常骨髓造血干/祖细胞集落形成受抑制,并上调造血干/祖细胞Fas表达。
Objective To observe the influence of negative factors of hematopoietic regulation in serum of normal bone marrow hematopoietic cells in patients with benzene poisoning aplastic anemia (BPAA). Methods The BPAA patients were selected as experimental subjects. Bone marrow cells from healthy volunteers were used to count the bone marrow hematopoietic stem / progenitor colony forming units (CFU). The changes of CD34 + cell rate and Fas expression were detected by flow cytometry Serum levels of IFN-γ, TNF-α and IL-2 were detected by enzyme linked immunosorbent assay (ELISA). Results The numbers of CFU-GM and CFU-E in bone marrow of BPAA patients were significantly decreased (all P <0.05), the percentage of CD34 + The expression of Fas in cells increased significantly (all P <0.05). The levels of IFN-γ, TNF-α and IL-2 in serum were also significantly increased (all P <0.05). Conclusions The level of hematopoietic negative regulatory factor in BPAA patients is elevated, which inhibits the colony formation of normal bone marrow hematopoietic stem / progenitor cells and up-regulates Fas expression in hematopoietic stem / progenitor cells.
