目的:探讨不同浓度枸杞叶(LBL)含药血清对长波紫外线(UVA)辐射致人真皮成纤维细胞(HSF)损伤的防护机制方法采用强度10J2cm -2的UVA照射损伤HSF造模,实验分为空白组、对照组、UVA模型组、空白血清组、UVA +高剂量LBL组、UVA +低剂量LBL组和UVA +VitC组。显微镜观察HSF形态,用MTT法检测细胞增殖活性,衰老相关β-半乳糖苷酶(SA-β-Gal)染色法计算衰老细胞率结果与空白组比较,UVA照射HSF损伤法可造成细胞增殖活性降低,衰老率增高(P均<0.01);与UVA+空白组比较,各含药血清组细胞活性增高,细胞衰老率降低(P均<0.01)。结论: LBL通过提高细胞增殖活性,降低衰老率发挥其保护HSF光损伤的机制。“,”objective to explore the different concentration of Chinese wolfberry leaves (LBL) medicated serum of long-wave ultraviolet radiation (UVA) human dermal fibroblasts (HSF) damage mechanism of protective methods the strength ten j. cm-2 HSF UVA irradiation damage mode, the experiment is divided into blank group, control group, UVA model group, blank serum group, the UVA + high-dose LBL group, low dose UVA + LBL group and UVA + VitC group.Microscope HSF form, with a determined by MTT method to detect cell proliferation activity, age-related beta galactose glucoside enzyme (SA-beta Gal) staining method calculation results compared with the blank group, senescent cel s rate HSF UVA irradiation damage redu ction method can cause cell proliferation activity, aging rate increased (P< 0.01);Compared with UVA +blank group, the medicated serum group increased cell activity, cel aging rate reduced (P<0.01).Conclusion:the LBL by increasing cel proliferation activity, decrease the rate of aging play its protection mechanism of HSF light damage.
