目的 探讨弹性蛋白酶在球囊损伤后动脉中的表达。方法 2F球囊导管损伤Wistar大鼠主动脉。用大鼠胰腺丝氨酸弹性蛋白酶Ⅱ的RNA探针进行原位杂交和Northern印迹杂交、抗弹性蛋白酶抗体进行免疫组化、胰腺弹性蛋白酶底物测定其活性 ;电镜观察迁徙细胞与其周围弹性纤维的连接处的数量。结果 球囊损伤后 ,迁徙细胞弹性蛋白酶mRNA、蛋白表达及其活性均有明显增加 ,细胞周围弹性纤维的连接处数量显著减少。结论 弹性蛋白酶是降解细胞外基质和动脉血管内弹性板、促使平滑肌细胞向内膜迁徙的重要原因。
Objective To investigate the expression of elastase in arteries after balloon injury. Method 2F balloon catheter injury Wistar rats aorta. In situ hybridization and Northern blot hybridization were performed with the RNA probe of rat pancreatic serine elastase II, anti-elastase antibody was used for immunohistochemistry and pancreatic elastase substrate was used to measure the activity. The junction between migratory cells and surrounding elastic fibers was observed under electron microscope quantity. Results After balloon injury, the mRNA and protein expression of migratory cell elastase and their activity were significantly increased, and the number of elastic fibers around the cells was significantly reduced. Conclusion Elastase is an important reason to degrade the extracellular matrix and the intra-arterial elastic plate and promote the migration of smooth muscle cells to the intima.
