目的:探讨梗阻性无精子症患者的附睾组织中附睾蛋白P34h mRNA及蛋白表达情况,并探讨其在梗阻性无精子症患者中的意义。方法:采用逆转录聚合酶链反应(RT-PCR)检测30例因不同梗阻(输精管或射精管水平梗阻)所致的梗阻性无精子症患者附睾组织中P34h基因mRNA的表达水平,Western Blot检测2组P34h蛋白的表达。结果:2组均有P34h mRNA的表达,输精管梗阻的P34h/β-actin均值为0.418±0.045,射精管梗阻的P34h/β-actin均值为0.719±0.069,输精管梗阻的附睾组织中P34h mRNA水平显著低于射精管梗阻(P<0.05)。2组亦均有P34h蛋白的表达,输精管梗阻平均光密度为0.095±0.014,射精管梗阻为0.276±0.054(P<0.05),两组P34h蛋白的表达差异有统计学意义。结论:P34h mRNA及其蛋白在不同梗阻因素所致的无精子症患者的附睾中表达水平有明显差异,提示P34h可能是梗阻性无精子症时导致精子成熟障碍的一个重要因素。
Objective: To investigate the expression of P34h mRNA and protein in the epididymis of patients with obstructive azoospermia, and to explore its significance in patients with obstructive azoospermia. Methods: The mRNA expression of P34h in the epididymis of 30 patients with obstructive azoospermia due to obstruction (vas deferens or ejaculation level obstruction) was detected by reverse transcription-polymerase chain reaction (RT-PCR). Western Blot 2 group P34h protein expression. Results: The expression of P34h mRNA in both groups was significantly lower than that in the control group (P <0.05). The mean P34h / β-actin in vas deferens obstruction was 0.418 ± 0.045. The mean P34h / β-actin in ejaculatory duct obstruction was 0.719 ± 0.069. Lower than ejaculatory duct obstruction (P <0.05). The expression of P34h protein was also found in both groups. The average optical density of vas deferens obstruction was 0.095 ± 0.014, and the ejaculatory duct obstruction was 0.276 ± 0.054 (P <0.05). There was significant difference in P34h protein expression between the two groups. Conclusion: The expression of P34h mRNA and protein in the epididymis of azoospermia patients with different obstructive factors are significantly different, suggesting that P34h may be an important factor in obstructive azoospermia resulting in sperm maturation disorders.
