目的 探讨白细胞介素 12 (IL- 12 )对哮喘患者辅助性 T细胞亚群的调节作用 .方法 收集 14例支气管哮喘患者 ,10例正常对照者的外周血单个核细胞 (PBMC) ,加入不同浓度 IL- 12进行体外培养 ,收集细胞培养上清液 ,以酶联免疫吸附法测定 IL - 4及 IFN-γ.并用酶联免疫斑点法(EL ISPOT)检测哮喘患者体外培养的 PBMC中的 Th1 和Th2 阳性率 .结果 哮喘患者 PBMC培养上清液中 IFN-γ产生减少 (P<0 .0 1) ,IL- 4产生增高 (P<0 .0 1) .IL- 12对 IFN-γ的产生有刺激作用 (P<0 .0 1) .较高浓度 IL - 12 (5μg·L- 1 )刺激 PBMC产生 IL - 4(P<0 .0 1) ,低浓度 IL - 12 (0 .0 5μg· L- 1 )抑制 IL- 4的产生 (P<0 .0 1) .EL ISPOT检测出的结果是 ,哮喘患者 PBMC中 Th1 阳性率与正常对照组无显著差别 (P>0 .0 5 ) ,而 Th2 阳性率明显增高 (P<0 .0 1) .不同浓度 IL- 12均能显著增高 Th1 的阳性率 (P<0 .0 1) ,较高浓度IL - 12亦能增高 Th2 阳性率 (P<0 .0 5 ) ,低浓度 IL - 12降低Th2 阳性率 (P<0 .0 1) .结论 IL- 12参与了哮喘的发病过程 ,它通过调节 Th1 / Th2 的活性及细胞因子的产生在哮喘的发病过程中发挥作用
Objective To investigate the regulatory effect of interleukin-12 (IL-12) on helper T cell subsets in asthmatic patients.Methods Peripheral blood mononuclear cells (PBMCs) were collected from 14 patients with bronchial asthma and 10 normal controls, IL-12 were cultured in vitro and the cell culture supernatants were harvested for determination of IL-4 and IFN-γ by enzyme-linked immunosorbent assay (ELISA). Th1 and Th2 positive rate.Results The production of IFN-γ in PBMC culture supernatant of asthmatic patients decreased (P <0.01) and the production of IL-4 increased (P <0.01) (P <0.01). IL - 4 (P <0.01) was stimulated by IL - 12 at a higher concentration (5μg · L -1) · L-1) inhibited the production of IL-4 (P <0.01). The results of EL ISPOT showed that the positive rate of Th1 in PBMC of asthmatic patients was not significantly different from the normal control group (P> 0.05) , While the positive rate of Th2 was significantly increased (P <0.01) .The IL-12 of different concentrations could significantly increase the positive rate of Th1 (P <0.01), while the higher concentration of IL-12 could also increase the positive rate of Th2 (P <0 .0 5 ), Low concentration of IL - 12 decreased the positive rate of Th2 (P <0.01) .Conclusion IL - 12 is involved in the pathogenesis of asthma by regulating Th1 / Th2 activity and cytokine production in the pathogenesis of asthma Play a role
