武汉地区测定柑桔抗寒性应在十一月中旬至翌年二月底进行。试材以春梢(即经过约八个月生长的正常枝梢)最具代表性。用其浸出液的电解质%计算,可以了解各品种在不同年份相对时期里的抗寒性。试材的低温处理应选择超过一部分品种受冻的临界低温,处理时间应当适当延长,其结果才能考验出各品种细胞膜的忍耐程度。电导法在选育抗寒性早期鉴定中具有一定的作用。计量耐寒性单位用简单回归法的b值比较要比用电解质渗出%更为准确。
