目的分析醉香含笑心材提取物(extract of trunk of Michelia macclurei Dandy.,ETMMD)的挥发性成分,并考察其对人乳腺癌细胞MDA-MB-231的体外生长抑制作用及诱导其凋亡的作用。方法水蒸气蒸馏法提取醉香含笑心材的挥发性成分,用气-质联用(GC-MS)技术进行分析,并用峰面积归一化法测定各成分相对含量;采用MTT法及流式细胞术分别检测6.25,12.50,25.00μg·mL-1的ETMMD对MDA-MB-231细胞体外生长抑制作用及诱导其凋亡的作用。结果共检出42个色谱峰,确定了其中41种化合物,占该挥发性成分总量的99.73%。ETMMD对MDA-MB-231细胞的抑制率呈浓度依赖关系。结论 ETMMD的挥发性成分以倍半萜烯及不饱和酮、不饱和酯类为主,此外还包括高级脂肪酸酯,以及一些甾体类化合物。ETMMD能明显抑制MDA-MB-231细胞的生长。浓度为6.25,12.50,25.00μg·mL-1的ETMMD可明显诱导人乳腺癌细胞MDA-MB-231的凋亡。
Objective To analyze the volatile components of extract of trunk of Michelia macclurei Dandy. (ETMMD) and investigate its inhibitory effect on the in vitro growth and apoptosis of human breast cancer cell line MDA-MB-231. . Methods The volatile components of L. lyophila L. with heartwood were extracted by steam distillation and analyzed by gas chromatography-mass spectrometry (GC-MS). The relative content of each component was determined by peak area normalization method. The MTT method and flow cytometry were used. The effects of ETMMD of 6.25, 12.50, and 25.00 μg·mL-1 on the growth inhibition and apoptosis of MDA-MB-231 cells in vitro were detected. Results A total of 42 peaks were detected and 41 compounds were identified, accounting for 99.73% of the total volatile components. The inhibitory rate of ETMMD on MDA-MB-231 cells was concentration-dependent. Conclusion The volatile components of ETMMD are mainly sesquiterpenes, unsaturated ketones and unsaturated esters. In addition, they also include higher fatty acid esters and some steroids. ETMMD can significantly inhibit the growth of MDA-MB-231 cells. The concentration of 6.25, 12.50, and 25.00 μg·mL-1 of ETMMD could significantly induce apoptosis of human breast cancer cell line MDA-MB-231.
