目的研究大孔吸附树脂分离纯化川明参茎叶中总黄酮的工艺条件及参数。方法以川明参茎叶中总黄酮的吸附量和解吸率为指标筛选大孔吸附树脂,研究所选树脂对川明参茎叶中总黄酮的吸附和解吸特性,并优化纯化的条件。结果AB-8型大孔树脂对川明参茎叶中总黄酮的静态饱和吸附量为50.52mg·g-1、解析率大于70%,动态吸附量为20.76mg·g-1、解析率86%;上柱前初提物中川明参总黄酮的含量为11.7%,经大孔吸附树脂分离纯化后产品中的总黄酮含量达26.74%,提高了2.29倍;总黄酮含量与抗氧化活性成正比。结论AB-8型大孔吸附树脂对川明参茎叶中的总黄酮有较好的分离纯化效果,总黄酮是川明参茎叶中的主要抗氧化活性成分。
Objective To study the conditions and parameters for the separation and purification of total flavonoids from stems and leaves of Panax chuanxiong by macroporous adsorption resin. Methods The adsorption and desorption of total flavonoids in stems and leaves of P. chuanxiong were used to screen macroporous resin, and the adsorption and desorption characteristics of total flavonoids in stems and leaves of P. chuanxiong stems were studied. The conditions for purification were optimized. Results The AB-8 macroporous resin had a static saturation adsorption amount of 50.52 mg·g-1 and a resolution of more than 70% for total flavonoids in stems and leaves of Schisandra chuanxiong. The dynamic adsorption capacity was 20.76 mg·g-1 and the resolution was 86. %; The content of total flavonoids in Chuanmei ginseng was 11.7% before the pre-column extraction. The content of total flavonoids in the product after separation and purification by macroporous adsorption resin reached 26.74%, increased by 2.29 times; the total flavonoid content and antioxidant activity became Proportional. Conclusion The AB-8 macroporous resin can effectively purify the total flavonoids from the stems and leaves of Chuanming ginseng. The total flavonoids are the main antioxidants in the stems and leaves of Chuanmin ginseng.
