目的 :探讨紧密连接蛋白occludin表达与人肝细胞癌发生和发展的关系。方法 :运用免疫组化技术检测occludin在人肝细胞癌中的表达 ,并进行苏木精 伊红 (HE)染色观察细胞结构。结果 :occludin主要定位于肝细胞膜 ,且癌旁肝组织的表达阳性率明显高于肝细胞癌 (P <0 .0 1)。癌旁肝组织occludin表达 ~ 级强度显著高于肝细胞癌 (P <0 .0 0 5 )。occludin在10例分化较好 (Ⅰ、Ⅱ级 )的肝细胞癌及其癌旁组织中的表达 ,其阳性率差异无显著性 (P >0 .0 5 ) ,但 33例分化较差 (Ⅲ、Ⅳ级 )的肝细胞癌中 ,occludin在癌旁组织中表达的阳性率高于癌组织 (P <0 .0 5 )。结论 :肝细胞癌与癌旁肝组织相比 ,occludin表达阳性率及其强度均明显下降 ,其改变在肝细胞癌的发生发展中可能起重要作用。
Objective: To investigate the expression of tight junction protein occludin and the occurrence and development of human hepatocellular carcinoma. Methods: The expression of occludin in human hepatocellular carcinoma was detected by immunohistochemistry and the cell structure was observed by hematoxylin and eosin (HE) staining. Results: Occludin mainly located in the liver cell membrane, and the positive expression rate of adjacent liver tissue was significantly higher than that of hepatocellular carcinoma (P <0.01). The expression level of occludin in adjacent noncancerous liver tissue was significantly higher than that in hepatocellular carcinoma (P <0.05). The positive rate of occludin in 10 cases of well-differentiated hepatocellular carcinoma (HCC) and its adjacent tissues was not significantly different (P> 0.05), but 33 cases had poor differentiation , Grade Ⅳ), the positive rate of occludin expression in adjacent tissues was higher than that in cancer tissues (P <0.05). Conclusion: The positive rate and the intensity of occludin expression in hepatocellular carcinoma are significantly lower than those in para-cancerous liver tissue. The changes may play an important role in the development of hepatocellular carcinoma.
