用细胞因子活性检测,Northernblot方法,在体外研究了抗细菌核心糖脂域McAb(EL1、EL3和3H4)对LPS诱导人外周血单核细胞(hPBMC)释放TNF-α和IL-6及其mRNA表达水平的影响。结果表明,EL1、EL3和3H4在体外有抑制LPS诱导细胞释放TNF-α和IL-6的作用;Northernblot结果证实,这3株McAb能分别降低细胞TNF-α和IL-6mRNA表达的水平;提示抗细菌核心糖脂域McAb可能通过与LPS的结合而中和LPS,从而降低或阻碍了效应细胞炎性细胞因子mRNA的表达,达到降低相应细胞因子的作用。
Cytokine activity assay and Northern blot method were used to study the effects of antibacterial core glycolipid McAbs (EL1, EL3 and 3H4) on the release of TNF-α, IL-6 and its mRNA from human peripheral blood mononuclear cells (hPBMC) induced by LPS Effect of expression level.
