目的:观察中药益肾助孕颗粒对围绝经期大鼠卵巢血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法:通过自然老化法构建围绝经期大鼠模型,选择处于围绝经期的大鼠(12~14月龄)40只,随机分为老年大鼠对照组、益肾助孕颗粒高、中、低剂量组。每组10只,分别用生理盐水2 mL灌胃、益肾助孕颗粒按等效剂量高剂量10 g.kg-1.d-1,中剂量5 g.kg-1.d-1,低剂量为2.5 g.kg-1.d-1,灌胃0.2 mL.g-1.d-1灌胃4周;另取5月龄大鼠10只作为青年大鼠对照组,生理盐水2 mL灌胃4周。免疫印迹法定量检测各组大鼠卵巢VEGF蛋白质的表达;RT-PCR检测各组大鼠卵巢VEGF mRNA的表达。结果:治疗前,与青年大鼠组VEGF蛋白、VEGF mRNA比较,老年对照组和研究组大鼠卵巢的VEGF蛋白、VEGF mRNA都表达明显高于青年组,差异显著,有统计学意义(P<0.05),用药4周后,与老年对照组大鼠卵巢VEGF蛋白、VEGF mRNA表达比,研究组大鼠卵巢VEGF蛋白、VEGF mRNA表达明显下降,差异显著,有统计学意义(P<0.05),并存在剂量依赖性。结论:益肾助孕颗粒能够下调围绝经期大鼠卵巢VEGF蛋白、VEGF mRNA的表达,可能通过调节VEGF治疗围绝经期综合征。
Objective: To observe the effect of Yishen Zhuyun Granule on the expression of vascular endothelial growth factor (VEGF) in perimenopausal rats. Methods: Perimenopausal rat models were established by natural aging method. Forty rats in the perimenopausal period (12 to 14 months old) were randomly divided into control group, Low dose group. 10 rats in each group were given gavage with 2 mL normal saline. The kidney-invigorating kidney-invigorating granules were treated with high dose of 10 g.kg-1.d-1 and low dose of 5 g.kg-1.d-1 A dose of 2.5 g.kg-1.d-1, gavage 0.2 mL.g-1.d-1 gavage for 4 weeks; another 5-month-old rats as a control group of young rats, saline 2 mL Gavage for 4 weeks. Immunohistochemistry was used to detect the expression of VEGF protein in ovaries of each group. The mRNA expression of VEGF in ovaries was detected by RT-PCR. Results: Compared with young rats, VEGF protein and VEGF mRNA in the old control group and the study group were significantly higher than those in the young group before treatment (P < 0.05). After 4 weeks of treatment, the expression of VEGF protein and VEGF mRNA in the ovary of the rats in the control group were significantly lower than those in the control group (P <0.05). The expressions of VEGF protein and VEGF mRNA in the ovary of the study group were significantly decreased And there is a dose-dependent. Conclusion: Yishen Zhuyun granule can down regulate the expression of VEGF protein and VEGF mRNA in the ovaries of perimenopausal rats, which may be related to the treatment of perimenopausal syndrome by regulating VEGF.
