目的探讨紫外线对系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMC)表达白介素-6(IL-6)、白介素-10(IL-10)mRNA的影响。方法提取SLE患者及健康对照的PBMC,用不同剂量311nm窄谱中波紫外线(NB-UVB)照射,以实时荧光相对定量聚合酶链反应分析照射前后PBMC IL-6、IL-10mRNA的相对表达。结果 SLE患者中狼疮肾炎(LN)患者PBMC高表达IL-6、IL-10mRNA,与非狼疮肾炎(非LN)患者及正常人差异很大,分别是正常人的(105.44±112.94)倍和(1.82±3.41)倍、(28.41±21.14)倍和(1.17±1.43)倍,其中狼疮肾炎(LN)患者PBMC高表达IL-6、IL-10 mRNA,且与临床中24h尿蛋白定量高度相关(r=0.82,0.87,P<0.05),而非狼疮肾炎系统性红斑狼疮患者与正常人相比,差异无统计学意义(t’=0.43,0.87,P>0.05);在紫外线照射后LN患者PBMC IL-6、IL-10mRNA的表达有升高也有降低,升高组SLE疾病活动指数、抗SSA抗体阳性率以及光敏性发生率明显高于降低组。结论紫外线可改变SLE患者PBMC表达IL-6、IL-10mRNA,紫外线可能通过此途径影响SLE病情。
Objective To investigate the effects of ultraviolet light on interleukin-6 (IL-6), interleukin-10 (IL-10) mRNA expression in peripheral blood mononuclear cells (PBMCs) from patients with systemic lupus erythematosus (SLE) Methods The PBMC of SLE patients and healthy controls were extracted and irradiated with different doses of NB-UVB at 311nm. The relative expression of IL-6 and IL-10mRNA in PBMCs before and after irradiation were analyzed by real-time fluorescence quantitative polymerase chain reaction. Results The levels of IL-6 and IL-10 mRNA in peripheral blood of patients with lupus nephritis (LN) were significantly different from those of non-lupus nephritis patients and non-LN patients, which were 105.44 ± 112.94 times and (1.82 ± 3.41) fold, (28.41 ± 21.14) fold and (1.17 ± 1.43) fold, respectively. IL-6 and IL-10 mRNA were highly expressed in PBMC of patients with lupus nephritis (LN) r = 0.82,0.87, P <0.05), while there was no significant difference between the patients with lupus nephritis and normal controls (t ’= 0.43,0.87, P> 0.05) The expression of IL-6 and IL-10mRNA in PBMCs increased and decreased, while the activity index of SLE disease, the positive rate of anti-SSA antibody and the photosensitivity in the elevated group were significantly higher than those in the reduced group. Conclusion UV can change the expression of IL-6 and IL-10 mRNA in peripheral blood mononuclear cells of SLE patients. Ultraviolet light may affect SLE in this way.
