AFLP是以PCR扩增为基础的,是扩增片段长度多态性的简称。它是RFLP和PCR相结合的一种标记技术,其基本原理是对基因组DNA限制性酶切片断的选择性扩增。AFLP的基本反应过程包括模板DNA制备、酶切片段的扩增和扩增产物的凝胶电泳分析3个步聚。AFLP是一种新颖和强大的DNA指纹技术,结合了RFLP和RAPD的优点,具有高效、快捷、稳定、可靠的特点。本文将AFLP和RFLP、RAPD、SSR等分子标记进行了比较,结果表明它可产生丰富而稳定的遗传标记,因而在林木分类、品种鉴定、遗传图谱构建、育种和目的基因定位等研究上有良好的应用前景。
AFLP is based on PCR amplification and is short for amplified fragment length polymorphism. It is a combination of RFLP and PCR labeling technology, the basic principle is the selective amplification of restriction fragments of genomic DNA. The basic reaction process of AFLP includes 3 steps of template DNA preparation, amplification of digested fragments and gel electrophoresis of amplification products. AFLP is a novel and powerful DNA fingerprinting technology that combines the advantages of RFLP and RAPD with the advantages of efficiency, speed, stability, and reliability. In this paper, AFLP and RFLP, RAPD, SSR and other molecular markers were compared, the results showed that it can produce rich and stable genetic markers, and thus in the classification of forest, identification of varieties, genetic map construction, breeding and gene mapping and other studies have good The application prospects.
