目的评价DWI动态监测新型血管阻断剂M410治疗兔VX2种植性肝癌的效果。方法建立28只兔VX2种植性肝癌模型,随机等分为2组,分别静脉注射M410(实验组)和生理盐水(对照组)。两组于给药前、给药后4h、1天、4天、7天、14天分别行MR平扫及DWI检查,并于各时间点取肿瘤组织行HE染色和CD34免疫组化检查,观察肿瘤体积、肿瘤整体及实质平均ADC值及病理变化。结果实验组肿瘤体积较对照组增长缓慢,给药后4~14天各时间点两组肿瘤体积差异均有统计学意义(P均<0.05)。肿瘤整体及实质平均ADC值:给药后1天实验组低于对照组,4天实验组高于对照组,差异均有统计学意义(P<0.05)。ADC值改变与肿瘤组织病理学结果相符。结论 DWI可动态反映新型血管阻断剂M410治疗兔VX2种植性肝癌后肿瘤分子水平的变化,通过测量肿瘤ADC值可早期监测M410的疗效。
Objective To evaluate the efficacy of DWI in monitoring the VX2 implanted hepatocellular carcinoma in rabbits with a new vascular blocker M410. Methods A total of 28 rabbit models of VX2 implanted hepatocellular carcinoma were established and randomly divided into 2 groups: M410 (experimental group) and normal saline (control group). The MR and DWI examinations were performed before and 4h, 1d, 4d, 7d and 14d after administration. The tumor tissues were harvested at each time point for HE staining and CD34 immunohistochemistry. Tumor volume, tumor overall and real average ADC values and pathological changes were observed. Results The tumor volume of the experimental group grew more slowly than that of the control group. The difference of tumor volume between the two groups was statistically significant at 4 to 14 days after administration (all P <0.05). Tumor overall and real average ADC values: one day after administration, the experimental group was lower than the control group, the experimental group on the 4th day was higher than the control group, the difference was statistically significant (P <0.05). ADC value changes consistent with histopathological findings. Conclusion DWI can dynamically reflect the change of tumor molecular level after the new vascular blocker M410 is used to treat VX2 inoculated with HCC. The curative effect of M410 can be monitored early by measuring the ADC value of the tumor.
