目的 研究细胞间粘附分子 - 1(ICAM- 1)在乙型肝炎 (乙肝 )病毒清除及肝癌免疫机制中的意义及细胞因子对 ICAM- 1的调节作用。方法 利用免疫组化技术对 HBV相关肝病肝组织ICAM- 1表达进行检测 ,同时检测乙肝组织内 HBc Ag表达状况。利用细胞酶联免疫技术观察 IFN-r、IFN- α、TNF对人肝癌细胞 H- 740 2 ICAM- 1表达的调节作用。结果 在慢性乙肝中 ,ICAM- 1表达愈强 ,肝内 HBc Ag表达愈少 ;肝癌 ICAM- 1表达较癌旁及肝硬化组织减弱 ;细胞因子对 ICAM- 1表达有增强作用。结论 慢性乙肝组织 ICAM- 1一定程度的有利于 HBV清除 ;ICAM- 1表达减少可能是肝癌发生过程中免疫逃逸的重要机制 ;IFN- r、IFN- α、TNF对 ICAM- 1表达有增强作用。
Objective To investigate the significance of intercellular adhesion molecule - 1 (ICAM - 1) in the clearance of hepatitis B virus (HBV) and the mechanism of liver cancer immunity and the regulation of ICAM - 1 by cytokines. Methods The expression of ICAM-1 in liver tissue of HBV-related liver disease was detected by immunohistochemistry and the expression of HBcAg in hepatitis B virus was detected. The effect of IFN-γ, IFN-α and TNF on the expression of ICAM-1 in HCC cell line H-7402 was observed by cell enzyme-linked immunosorbent assay. Results In chronic hepatitis B, the stronger the expression of ICAM-1, the less the expression of HBc Ag in liver. The expression of ICAM-1 in hepatocellular carcinoma was weaker than that in paracancer and cirrhosis. The expression of ICAM-1 was enhanced by cytokines. CONCLUSION: ICAM-1 in chronic hepatitis B is beneficial to HBV clearance to a certain extent. The decrease of ICAM-1 expression may be an important mechanism of immune escape in hepatocellular carcinoma. IFN-γ, IFN-α and TNF may enhance the expression of ICAM-1.
