海洋来源链霉菌M1O抗白色念珠菌活性物质研究

对从大连海域沉积物样品中分离得到的海洋放线菌进行活性筛选,获得了1株对白色念珠菌具有良好拮抗活性的菌株M10.基于16S rRNA的系统发育分析表明,M10菌株与分离自植物根际土壤的模式菌种摩洛哥链霉菌(Streptomyces marokkonensis)DSM 41918T的亲缘关系最近.对M10菌株的发酵产物进行活性追踪,M10菌株拮抗白色念珠菌的活性物质存在于乙酸乙酯相中,在低于60℃时具有较好的热稳定性,在pH值介于3~8之间具有良好的酸碱稳定性,但pH值升至9~12时活性明显下降.采用溶剂萃取、柱色谱层析及制备HPLC等方法对M10菌株发酵液中的有机相活性物质进行了分离,得到2种纯化合物A和B,其最小抑菌浓度分别为0.125、0.250 μg/cm3;红外光谱分析表明A和B组分可能是不饱和酮酯类和不饱和苯衍生物.本文首次报道了海洋来源的摩洛哥链霉菌近缘菌株抗白色念珠菌活性物质的分离纯化,可为进一步开发利用海洋来源的新型抗真菌药物库提供参考.
