目的研究低氧对肺动脉平滑肌细胞(pulmonary arterial smooth muscle cells,PASMCs)白细胞介素-1β、4、10、13(IL-1β、4、10、13)活性的影响,及复合培养下肺微血管内皮细胞(pulmonary microvascular endothelial cells,PMVECs)对其调控作用。方法大鼠PASMCs单独培养或与PMVECs复合培养,分为正常组、低氧作用2h(H2)、6h(H6)、12h(H12)、24h(H24)组,采用ELISA法测定各组培养细胞上清中IL-1β、IL-4、IL-10、IL-13的活性。结果低氧刺激后PASMCs的IL-1β、IL-4、IL-10、IL-13活性2h开始升高,6h达高峰,12h降低;各时相点复合培养组均低于对应的单独培养组(P<0.01)。结论低氧可以激活PASMCs的IL-1β、IL-4、IL-10、IL-13活性,在复合培养条件下,PMVECs对此效应具有下调性影响。
Objective To investigate the effects of hypoxia on the activities of interleukin-1β, 4,10,13 (IL-1β, 4,10,13) in pulmonary arterial smooth muscle cells (PASMCs) (Pulmonary microvascular endothelial cells, PMVECs) on its regulation. Methods PASMCs were cultured in vitro or in combination with PMVECs. The cells were divided into normal group, hypoxia for 2h (H2), 6h (H6), 12h (H12) and 24h (H24) Qing IL-1β, IL-4, IL-10, IL-13 activity. Results After hypoxia stimulation, the activities of IL-1β, IL-4, IL-10 and IL-13 in PASMCs increased at 2h, reached the peak at 6h and decreased at 12h, and were lower than those of the corresponding cultured groups (P <0.01). CONCLUSION: Hypoxia activates IL-1β, IL-4, IL-10 and IL-13 in PASMCs. PMVECs have a down-regulation effect on this effect under the condition of compound culture.
