目的 研究C-反应蛋白(C-reactive protein,CRP)对低氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)表达的影响。方法 利用COCl_2(浓度为100μmol/L)模拟缺氧环境,使其分别与4个试验组的CRP(浓度分别为5,10,50,100μg/ml)同时作用于脐静脉内皮细胞,以仅加CoCl_2(100μmol/L)组为对照;同时设空白对照组。刺激24h后提取蛋白。用Western-blotting半定量检测HIF-1α的蛋白表达量,数据用SPSS11.0软件进行统计分析。结果 CRP在5μg/ml水平即减少HIF-1α表达(P<0.001),在100μg/ml水平达到最大作用,其作用效果呈剂量反应关系(P<0.001)。结论 CRP抑制缺氧状态下人脐静脉内皮细胞表达HIF-1α。此结果证实CRP直接作用于血管,抑制缺氧组织血管新生。
Objective To investigate the effect of C-reactive protein (CRP) on the expression of hypoxia inducible factor-1α (HIF-1α). Methods COCl 2 (100 μmol / L) was used to simulate the hypoxia environment and the effects of COCl 2 (5, 10, 50 and 100 μg / ml, respectively) on umbilical vein endothelial cells (100μmol / L) group as a control; also set the blank control group. Protein was extracted 24h after stimulation. Western-blotting semi-quantitative detection of HIF-1α protein expression data using SPSS11.0 software for statistical analysis. Results CRP decreased the expression of HIF-1α (P <0.001) at 5μg / ml and reached the maximum at 100μg / ml. The effect was dose-response (P <0.001). Conclusion CRP inhibits HIF-1α expression in human umbilical vein endothelial cells under hypoxia. This result confirms that CRP acts directly on blood vessels and inhibits angiogenesis in hypoxic tissues.
