目的 研究MTT法检测癌细胞对抗癌药物的敏感性与临床个体化化疗的相关性。方法 取 5 7例癌症患者新鲜癌细胞 ,采用MTT法检测癌细胞对 8种常用抗癌药的敏感性 ,成功 46例 ,根据药敏结果对患者进行化疗 ,无敏感药物者按经验方案化疗 ,观察实验与临床结果的相关性。结果 药敏试验阳性者 2 5例 ,阴性者 2 1例。实验提示敏感药物化疗者的有效率为 76 .0 %(19/2 5 ) ,耐药者按经验方案化疗 ,其有效率仅为 2 8.6 % (6 /2 1) ,两者有非常显著性差异 ,P <0 .0 1。实验与临床符合率达 73 .9%(34/46 ) ;说明MTT法对癌症患者临床个体化化疗效果有预见价值。结论 MTT法体外癌细胞药敏试验可以指导临床个体化治疗。
Objective To study the correlation between the sensitivity of anti-cancer drugs detected by MTT assay and clinical individualized chemotherapy. Methods A total of 57 fresh cancer cells from cancer patients were used. MTT assay was used to detect the sensitivity of cancer cells to 8 commonly used anticancer drugs. 46 patients were successfully treated. Chemosensitivity was used to administer chemotherapy to patients, and no drug-sensitive patients were treated by experience. Observe the correlation between the experimental and clinical results. Results There were 25 patients with positive drug sensitivity test and 21 patients with negative drug sensitivity test. The experimental results showed that the effective rate of chemotherapy for sensitive drugs was 76.0% (19/2 5 ), and those with drug resistance were treated according to empirical protocols. The effective rate was only 28.6% (6/21), and the two were very significant. Difference, P <0. 0 1. The coincidence rate between the experiment and the clinic was 73.9% (34/46), indicating that the MTT assay has predictive value for the clinical individualized chemotherapy effect of cancer patients. Conclusion The in vitro cancer susceptibility test of MTT can guide individualized clinical treatment.
