目的探讨不同浓度西罗莫司对急性髓系白血病细胞株Kasumi-1增殖、凋亡和细胞周期的影响。方法 MTT法检测西罗莫司对Kasumi-1细胞增殖的影响,流式细胞术检测西罗莫司对Kasumi-1细胞凋亡和细胞周期的变化,免疫荧光法观察西罗莫司处理前后Kasumi-1细胞核形态变化。结果不同浓度的西罗莫司均能抑制Kasumi-1细胞的增殖,促进其凋亡,并使细胞阻滞在G0/G1期。Kasumi-1细胞对西罗莫司表现为剂量依赖性,并在100 ng/mL左右达到最大效应。结论西罗莫司能够显著抑制Kasumi-1细胞增殖,并对凋亡起促进作用。
Objective To investigate the effects of different concentrations of sirolimus on proliferation, apoptosis and cell cycle of acute myeloid leukemia cell line Kasumi-1. Methods MTT assay was used to determine the effect of sirolimus on the proliferation of Kasumi-1 cells. The changes of apoptosis and cell cycle of Kasumi-1 cells were detected by flow cytometry. Kasumi -1 nuclear morphological changes. Results All concentrations of sirolimus inhibited the proliferation of Kasumi-1 cells, promoted the apoptosis and arrested cells in G0 / G1 phase. Kasumi-1 cells showed dose-dependent effects on sirolimus and achieved maximal effects at around 100 ng / mL. Conclusion Sirolimus can significantly inhibit the proliferation of Kasumi-1 cells and promote the apoptosis.
