本文对醛糖还原酶筛选模型进行了系统研究,考察了影响醛糖还原酶模型的主要因素:醛糖还原酶量、NADPH量、DL-甘油醇量和反应体系的温度。在已有单因素实验的基础上,设计正交实验进行综合研究。结果表明醛糖还原酶抑制剂筛选模型的最佳条件为:35℃的温度,醛糖还原酶量为120μl,NADPH量为30μl,DL-甘油醇量为4μl。
In this paper, the model of aldose reductase screening was systematically investigated. The main factors affecting aldose reductase model were investigated: the amount of aldose reductase, the amount of NADPH, the amount of DL-glycerol and the temperature of the reaction system. Based on the existing single-factor experiments, orthogonal experiments are designed to conduct a comprehensive study. The results showed that the optimum conditions of the aldose reductase inhibitor screening model were as follows: the temperature was 35 ℃, the amount of aldose reductase was 120μl, the amount of NADPH was 30μl and the amount of DL-glycerol was 4μl.
