目的检测强直性脊柱炎外周血单个核细胞中急性时相反应蛋白表达水平,探讨其与强直性脊柱炎发病的关系。方法收集10例强直性脊柱炎患者和9例健康对照,提取外周血单个核细胞,采用相对和绝对定量的等量异位标签串联质谱技术检测强直性脊柱炎患者外周血单个核细胞中的急性时相反应蛋白表达水平,并运用Western blot方法进行验证差异表达的触珠蛋白(Hp)和铜蓝蛋白(Cp)。结果基于质谱分析的蛋白组学显示,19种急性时相反应蛋白在强直性脊柱炎外周血单个核细胞中蛋白表达水平明显升高(P<0.05),主要包括补体C3、纤粘蛋白(FN1)、血浆酶原(PLG)、转铁蛋白(TF)、Hp和Cp;Western blot检测Hp和Cp的表达水平与相对和绝对定量的等量异位标签串联质谱技术检测结果具有相同的变化趋势。结论急性时相反应蛋白差异表达可能参与了强直性脊柱炎的发生、发展,有望作为强直性脊柱炎的临床生物标记物。
Objective To detect the expression of acute phase reaction protein in peripheral blood mononuclear cells of ankylosing spondylitis and to explore its relationship with the pathogenesis of ankylosing spondylitis. Methods Ten patients with ankylosing spondylitis and nine healthy controls were collected and peripheral blood mononuclear cells (PBMCs) were extracted. Relative and absolute quantitative isobaric label tandem mass spectrometry (MS / MS) was used to detect acute peripheral blood mononuclear cells in patients with ankylosing spondylitis Phase reaction protein expression level, and the use of Western blot method to verify differentially expressed haptoglobin (Hp) and ceruloplasmin (Cp). Results Proteomics analysis based on mass spectrometry showed that the protein expression of 19 kinds of acute phase reaction proteins in peripheral blood mononuclear cells of ankylosing spondylitis was significantly increased (P <0.05), mainly including complement C3, fibronectin (FN1 (PLG), transferrin (TF), Hp and Cp. Western blot showed that the expression of Hp and Cp had the same trend as that of relative and absolute quantification of isobaric label tandem mass spectrometry . Conclusion The differential expression of acute phase reaction protein may be involved in the occurrence and development of ankylosing spondylitis, which is expected to be an effective clinical biomarker for ankylosing spondylitis.
