目的制备TRAIL及其5种受体的特异性基因探针。方法以PHA刺激人外周血淋巴细胞(PBL)增殖,从而诱导TRAIL及其受体的mRNA转录水平提高,然后用RTPCR方法扩增出TRAIL及其5种受体的DNA基因片段,并采用基因测序法确证所得基因片段序列的正确性。结果获得了TRAIL及其5种受体的DNA基因片段。结论所用方法正确,为研究TRAIL及其受体调控细胞凋亡的机制提供了有价值的工具。
Objective To prepare specific gene probes of TRAIL and its five receptors. Methods The proliferation of human peripheral blood lymphocytes (PBLs) was stimulated with PHA to induce the mRNA transcription of TRAIL and its receptor. Then the DNA fragments of TRAIL and its five receptors were amplified by RTPCR and sequenced by gene sequencing Method to confirm the correctness of the obtained gene fragment sequence. Results The DNA fragments of TRAIL and its five receptors were obtained. Conclusion The method is correct, which provides a valuable tool for studying the mechanism of TRAIL and its receptor regulating apoptosis.
