近日,上海市农业科学院生物所在源于苹果的抗草甘膦基因的研究方面取得新进展,以院生物所姚泉洪研究员为通讯作者、田永生为第一作者的研究论文“Mutation by DNA shuffling of 5-enopyruvylshikimate-3-phosphate synthase from Malus domestica for improved glyphosate resistance”近日被WILEY数据库的植物生物技术著名学术期刊Plant biotechnology journa(l影响因子:6.279)接受。该文首先通过RACE技术从苹果(Malus domestica)中克隆了草甘膦靶酶(5-烯醇丙酮莽草酸-3-磷酸合成酶)基因(MdEPSPS),并通过基因重排技术对其进行了改造,获得了一个含有8个氨基酸位点突变的可以耐受60 mM草甘膦的突变基因;然后对其进行了定点回复突变及酶动力学参数测
