目的通过检测Rb、p16、p53、p19、p21在衰老大鼠睾丸组织中的表达,探讨何首乌饮抗大鼠睾丸组织衰老的机制。方法选用8周龄SD雄性大鼠84只,随机分为正常组、模型组、何首乌饮高、中、低剂量组、何首乌丸剂组、自然恢复组,每组12只,采用D-半乳糖连续腹腔注射建立亚急性衰老大鼠模型。采用免疫组织化学、Western blotting和RT-PCR法检测磷酸化Rb蛋白(pRb)、p16、p53、p19、p21在大鼠睾丸组织的表达差异。结果模型组大鼠睾丸组织Rb、p53、p16、p19、p21表达明显增加,pRb表达明显减少(P<0.05);何首乌饮、何首乌丸组能显著提高pRb的表达,降低Rb、p53、p16、p19、p21表达(P<0.05),何首乌饮高剂量组调节作用大于其他各组(P<0.05)。结论何首乌饮通过调节睾丸组织Rb/p53信号转导通路相关基因和蛋白表达起到延缓衰老的作用。
OBJECTIVE: To investigate the mechanism of Heshouwuyin on the senescence of testis in rats by detecting the expression of Rb, p16, p53, p19 and p21 in the testis of aging rats. Methods Eighty-four SD male rats aged 8 weeks were randomly divided into normal group, model group, Polygonum multiflorum high, medium and low dose groups, Hepteryx pill group and natural recovery group, with 12 rats in each group. Subacute injection to establish sub-acute aging rat model. The expression of phosphorylated Rb protein (pRb), p16, p53, p19 and p21 in testis tissue of rats were detected by immunohistochemistry, Western blotting and RT-PCR. Results The expression of Rb, p53, p16, p19 and p21 in testis tissue and the expression of pRb in testis tissue of model group were significantly increased (P <0.05) p19 and p21 (P <0.05), and the regulation effect of high dose group of Polygonum multiflorum was higher than other groups (P <0.05). Conclusion Polygonum multiflorum plays an anti-aging role by regulating the gene and protein expression of Rb / p53 signal transduction pathway in testicular tissue.
