六亚甲基二乙酰胺(HMBA)可以诱导鼠红白血病细胞合成血红蛋白,人白血病细胞趋于成熟及人实体瘤细胞向正常分化。但HMBA对人胃癌细胞表面抗原的影响则未见报道。作者应用HMBA体外诱导人胃癌细胞系LHA-GC-I、GGC及原代培养胃癌细胞,以抗胃癌单克隆抗体ID_(1),_(2)与癌细胞表面抗原结合,用免疫过氧化物酶染色和直接免疫荧光法检测。癌细胞经HMBA诱导120小时后,出现阴性结果的概率为0.78。说明胃癌细胞表面的癌相关抗原有丢失现象。这一初步结果为HMBA的临床应用提供了实验依据。
Hexamethylene bisacetamide (HMBA) can induce hemoglobin synthesis in murine erythroleukemia cells, human leukemic cells tend to mature and human solid tumor cells differentiate to normal. However, the effect of HMBA on the surface antigen of human gastric cancer cells has not been reported. The authors used HMBA to induce human gastric cancer cell lines LHA-GC-I, GGC and primary cultured gastric cancer cells in vitro. The anti-gastric cancer monoclonal antibodies ID_(1), _(2) were combined with the surface antigens of cancer cells, using immunoperoxides. Enzyme staining and direct immunofluorescence assay. After 120 hours of induction with HMBA, the probability of a negative result was 0.78. It shows that there is a loss of cancer-associated antigen on the surface of gastric cancer cells. This preliminary result provides experimental evidence for the clinical application of HMBA.
