目的:研究红芪多糖(HPS)对人牙周膜细胞(h PDLCs)增殖及分泌IL-1β、IL-6的影响。方法:采用改良组织块酶消化法培养人牙周膜细胞,经免疫组化SP法进行鉴定,MTT法检测细胞增殖情况,荧光实时定量PCR和酶联免疫法检测红芪多糖对LPS作用的h PDLCs分泌IL-1β、IL-6的影响。结果:10μg/m L HPS能够促进人牙周膜细胞增殖(P<0.05);10μg/m L LPS可显著刺激人牙周膜细胞分泌IL-1β、IL-6(P<0.01),而给予10μg/m L HPS能抑制LPS刺激人牙周膜细胞分泌IL-1β、IL-6,且与LPS组具有显著性差异(P<0.01)。结论:适宜浓度的红芪多糖能够促进体外培养的人牙周膜细胞增殖,并抑制LPS作用的人牙周膜细胞分泌IL-1β、IL-6。
Objective: To study the effects of HsPS on the proliferation and secretion of IL-1β and IL-6 in human PDLCs. Methods: Human periodontal ligament cells were cultured with modified tissue enzyme digestion method and identified by immunohistochemical SP method. Cell proliferation was detected by MTT assay. Real-time quantitative PCR and enzyme-linked immunosorbent assay were used to detect h PDLCs secrete IL-1β, IL-6. Results: 10μg / mL HPS could promote the proliferation of human periodontal ligament cells (P <0.05); 10μg / mL LPS could significantly stimulate the secretion of IL-1β and IL-6 in human periodontal ligament cells (P <0.01) HPS inhibited the LPS-induced secretion of IL-1β and IL-6 by human periodontal ligament cells with 10 μg / mL HPS, which was significantly different from LPS group (P <0.01). Conclusion: Hedysari polysaccharide can promote the proliferation of cultured human periodontal ligament cells in vitro and the secretion of IL-1β and IL-6 by LPS-treated human periodontal ligament cells.
