目的:建立柴芍肠宁胶囊中盐酸小檗碱的含量测定方法,考察不同煎煮方式对盐酸小檗碱含量的影响。方法:采用HPLC测定盐酸小檗碱含量,DIKMA ODS-C18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-0.1%磷酸(每100 mL加十二烷基磺酸钠0.1 g)(50∶50),检测波长345 nm。考察黄连与其他药味合煎或分煎时对盐酸小檗碱含量的影响。结果:盐酸小檗碱在2.4~24 mg·L-1线性关系良好(r=1),平均回收率101.14%,RSD 2.48%。分煎、合煎样品中盐酸小檗碱平均质量分数分别为0.128 2,0.078 6 mg·g-1,RSD依次为1.02%,1.25%。结论:黄连与其他药味分煎时盐酸小檗碱含量显著高于合煎,为柴芍肠宁胶囊的临床应用提供参考。
OBJECTIVE: To establish a method for the determination of berberine hydrochloride in Chai-shao-Chang-ning capsule and investigate the effects of different decoctions on the content of berberine hydrochloride. Methods: The content of berberine hydrochloride was determined by HPLC on a DIKMA ODS-C18 column (4.6 mm × 250 mm, 5 μm) with a mobile phase of acetonitrile-0.1% phosphoric acid (0.1 g per 100 mL plus sodium dodecyl sulfate) 50:50), detection wavelength 345 nm. To investigate the effects of Coptis chinensis and other herbs on the content of berberine hydrochloride when decocted or decocted. Results: Berberine hydrochloride had a good linear relationship (r = 1) at 2.4-24 mg · L -1 with an average recovery of 101.14% and a RSD of 2.48%. The average contents of berberine hydrochloride in the decoction samples and the decocted samples were 0.128 and 2.0.078 6 mg · g-1, respectively. The RSDs were 1.02% and 1.25%, respectively. Conclusion: The contents of berberine hydrochloride in Coptis chinensis and other medicinal herbs were significantly higher than those in Coptis chinensis decoction, which provided a reference for the clinical application of Chai-shan Chang-ning capsule.
