目的研究甲醛和甲苯联合染毒致小鼠脑组织氧化损伤作用,探讨其联合毒性效应的类型。方法采用静式吸入染毒,将小鼠暴露于不同浓度的甲醛、甲苯及两者的混合气体中。染毒结束后测定脑组织中活性氧(ROS)、谷胱甘肽(GSH)、丙二醛(MDA)、超氧化物歧化酶(SOD)和一氧化氮合酶(NOS)。结果甲醛和甲苯染毒可致小鼠脑组织ROS、MDA含量和NOS活力增高,SOD活力和GSH含量降低(P<0.01),其中联合组(5 mg/m3+2 000 mg/m3)的影响最为明显;二者联合染毒致小鼠脑组织氧化损伤存在交互作用(P<0.05),且表现为协同。结论甲醛和甲苯吸入可致小鼠脑组织氧化损伤,二者联合染毒具有一定的协同作用。
OBJECTIVE To study the oxidative injury of brain tissue of mice induced by formaldehyde and toluene and to explore the type of combined toxic effects. Methods The mice were exposed to different concentrations of formaldehyde, toluene and mixtures of both by inhalation. At the end of the treatment, reactive oxygen species (ROS), glutathione (GSH), malondialdehyde (MDA), superoxide dismutase (SOD) and nitric oxide synthase (NOS) were measured in brain tissue. Results Exposure of formaldehyde and toluene caused the increase of ROS, MDA and NOS activity, SOD activity and GSH content in brain tissue of mice (P <0.01), and the combination group (5 mg / m 3 + 2 000 mg / m 3) The most obvious; the combination of the two induced oxidative damage in brain tissue of mice interaction (P <0.05), and showed synergies. Conclusions Formaldehyde and toluene inhalation can cause oxidative damage in brain tissue of mice, and the synergistic effect between the two combined exposure.
