目的 观察三苯氧胺(Tamoxifen, TAM)及雌激素(E2)对乳腺癌MCF-7细胞株耐药基因mdr-1mRNA水平的影响;方法 应用RT-PCR法观察TAM及E2在不同时间、浓度作用下细胞内mdr-1mRNA水平的变化;结果TAM可以使MCF-7细胞内mdr-1mRNA水平降低,而E2则相反。且以上变化表现出剂量、时间依赖关系。结论TAM通过降低mdr-1mRNA水平,使肿瘤细胞的耐药性降低,有利于抗肿瘤药物更好的发挥作用。
Objective To observe the effect of tamoxifen (TAM) and estrogen (E2) on the mdr-1 mRNA level of breast cancer MCF-7 cell line; Methods RT-PCR method was used to observe the effects of TAM and E2 on the cell resistance of cells at different time and concentration. Changes in mdr-1 mRNA levels; results TAM can make mdr-1 mRNA levels in MCF-7 cells decreased, while E2 is the opposite. And the above changes show a dose-time dependent relationship. Conclusions TAM can reduce the resistance of tumor cells by decreasing the mdr-1 mRNA level, which is beneficial to the anti-cancer drugs.
