电磁脉冲对猴晶状体上皮细胞CyclinE表达的影响

目的 观察电磁脉冲(EMP)对猴晶状体上皮细胞Cyclin E表达水平的影响。方法 6×10~4 V/m场强的EMP辐射恒河猴30次,应用免疫组化、原位杂交和图像分析技术对晶状体上皮细胞Cyclin E蛋白及mRNA表达水平进行检测。结果 高场强EMP辐射干扰晶状体上皮细胞的正常增殖,照后30d,Cyclin E蛋白表达降低,其对细胞周期调控作用减弱。辐射损伤经修复后,照后136d和265d,Cyclin E蛋白表达水平升高,正性调控作用加强,晶状体上皮细胞增生活跃。蛋白与mRNA表达同步且一致。结论 EMP辐射影响细胞周期进程,上皮细胞增殖异常可能是导致白内障的重要原因。 Objective To observe the effect of electromagnetic pulse (EMP) on Cyclin E expression in lens epithelial cells of monkey. Methods Rhesus macaques were irradiated 30 times with EMP at a field intensity of 6 × 10 ~ 4 V / m. The expression of Cyclin E protein and mRNA in lens epithelial cells were detected by immunohistochemistry, in situ hybridization and image analysis. Results High-field intensity EMP radiation interfered the normal proliferation of lens epithelial cells. The expression of Cyclin E protein decreased 30 days after irradiation, and its effect on cell cycle was weakened. After the radiation injury was repaired, the expression of Cyclin E protein increased at 136 days and 265 days after irradiation, the positive regulatory effect was enhanced and the lens epithelial cells proliferated actively. Protein and mRNA expression are synchronized and consistent. Conclusion EMP radiation affects cell cycle progression and abnormal proliferation of epithelial cells may be an important cause of cataract.
