目的:探讨膜辅助蛋白(membrane cofactor protein,MCP)与宫颈癌细胞免疫逃逸之间的关系,揭示MCP在宫颈癌发生、发展中的作用。方法:以宫颈癌细胞系ME180、宫颈癌组织和癌旁正常组织为材料,采用real-time PCR和Western blot检测MCP基因mRNA和蛋白质表达水平;利用小干扰RNA(small interference RNA,siRNA)沉默MCP基因表达后,四甲基偶氮唑盐(MTT)比色法检查宫颈癌细胞的增殖,以Transwell评估宫颈癌细胞的迁移能力。结果:MCP的表达在宫颈癌组织中明显高于其配对的癌旁正常组织,抑制MCP基因的表达能显著降低ME180细胞的增殖、迁移能力。结论:MCP在宫颈癌细胞中的高表达可能是肿瘤细胞免疫逃逸的重要机制,在宫颈癌的发生发展中发挥重要作用。
Objective: To investigate the relationship between membrane cofactor protein (MCP) and immune escape of cervical cancer cells and to reveal the role of MCP in the occurrence and development of cervical cancer. Methods: The mRNA and protein expression levels of MCP gene were detected by real-time PCR and Western blot in cervical cancer cell line ME180, cervical cancer and adjacent normal tissues. MCP (Small Interfering RNA) was used to silence MCP After gene expression, MTT assay was used to examine the proliferation of cervical cancer cells. Transwell was used to assess the migration of cervical cancer cells. Results: The expression of MCP in cervical cancer was significantly higher than that in paired adjacent normal tissues. The inhibition of MCP gene expression could significantly reduce the proliferation and migration of ME180 cells. Conclusion: The high expression of MCP in cervical cancer cells may be an important mechanism of immune escape of tumor cells and play an important role in the occurrence and development of cervical cancer.
