本实验首次证明,用与河南林县食管癌高发相关的二乙基亚硝胺和N-3-甲基丁基-N-1-甲基丙酮基亚硝胺处理正常人胚肺成纤维样细胞,可使其发生不同程度的恶性转化(在半固体琼脂中生长,异种接种产生肿瘤)。转化细胞寿命明显延长。致癌物处理前先进行半固体琼脂预筛可富集易于转化的细胞,有助于建立人体细胞体外恶变系统。
This experiment demonstrated for the first time that normal human embryonic lung fibroblasts were treated with diethylnitrosamine and N-3-methylbutyl-N-1-methylacetonylnitrosamine associated with high incidence of esophageal cancer in Lin County, Henan Province. The cells can cause various degrees of malignant transformation (growth in semi-solid agar and inoculation of tumors). The life span of transformed cells is significantly longer. Semi-solid agar pre-screening prior to carcinogen treatment can enrich easily transformed cells and help establish a malignant system of human cells in vitro.
