目的:探讨CD4~+CD25~+调节性T细胞(Tregs)在2,4-二硝基氯苯(DNCB)和醋酸(AA)复合法诱发的小鼠溃疡性结肠炎中的变化规律及中药灌肠1号治疗的作用机制。方法:建立DNCB+AA复合法诱发的小鼠溃疡性结肠炎模型,设立正常对照、模型对照、柳氮磺氨吡啶(SASP)和中药灌肠1号治疗共4组,用流式细胞仪检测各组外周血、肠系膜淋巴结CD4~+CD25~+Tregs细胞的变化以及结肠组织病理学改变。结果:建模后1~2周,模型组外周血和肠系膜淋巴结的CD4~+CD25~+Tregs细胞数量较正常组明显下降,而使用SASP和中药灌肠Ⅰ号治疗后该细胞亚群明显高于模型组。结论:具有较强免疫抑制作用的CD4~+CD25~+Tregs细胞亚群在小鼠实验性溃疡性结肠炎的发病及病情转归中可能具有重要免疫调节作用,中药灌肠1号通过调节Tregs细胞的数量对溃疡性结肠炎发挥治疗作用。
Objective: To investigate the regularity of changes in CD4~+CD25~+ regulatory T cells (Tregs) induced by 2,4-dinitrochlorobenzene (DNCB) and acetic acid (AA) combined ulcerative colitis in mice and The mechanism of enema 1 treatment. METHODS: A rat model of ulcerative colitis induced by DNCB + AA complex method was established. Four groups of normal control, model control, sulfasalmide (SASP) and traditional Chinese medicine enema No. 1 treatment were established, and each group was examined by flow cytometry. Changes of CD4~+CD25~+Tregs cells in the peripheral blood and mesenteric lymph nodes and pathological changes in the colon. RESULTS: 1 to 2 weeks after modeling, the number of CD4~+CD25~+Tregs cells in the peripheral blood and mesenteric lymph nodes of the model group was significantly lower than that in the normal group, but after treatment with SASP and the Chinese medicine enema I, the subpopulation was significantly higher Model group. CONCLUSION: The CD4~+CD25~+Tregs subpopulations with strong immunosuppressive effects may have important immunoregulatory effects in the pathogenesis and prognosis of experimental ulcerative colitis in mice. Chinese medicine enema No.1 regulates Tregs cells. The amount of ulcerative colitis plays a therapeutic role.
