甘薯经过酶解、热水浸提、除蛋白、透析、乙醇沉淀,得到甘薯多糖。通过SDS-PAGE电泳方法,分离得到4种纯多糖组分。利用纤维素酶水解甘薯多糖,显著提高了可溶性多糖的产量。对不同条件下甘薯多糖酶法提取的影响因素进行了研究,结果表明,利用酶法结合热水浸提甘薯多糖,提取率达到5.55%。通过正交试验得到酶解最适作用条件为:反应时间90min、纤维素酶添加量0.04%(酶活性≧10000μg/g)、反应温度30℃。
Sweet potato after enzymolysis, hot water extraction, in addition to protein, dialysis, ethanol precipitation, to give sweet potato polysaccharides. By SDS-PAGE electrophoresis method, isolated four pure polysaccharide components. The hydrolysis of sweet potato polysaccharides by cellulase significantly increased the yield of soluble polysaccharides. The factors affecting the enzymatic extraction of sweet potato polysaccharides under different conditions were studied. The results showed that the extraction rate of sweet potato polysaccharides was 5.55% by enzymatic method combined with hot water. The optimum conditions for enzymolysis obtained by orthogonal test were: reaction time 90min, cellulase 0.04% (enzyme activity ≧ 10000μg / g), reaction temperature 30 ℃.
