目的研究猪链球菌2型(SS2)感染对TGFβ细胞凋亡蛋白表达的影响,为疫苗研发和药物救治提供参考。方法将30只清洁级昆明小鼠随机分为对照组、轻度感染组及重度感染组各10只;轻度和重度感染组小鼠分别腹腔注射猪链球菌2型2×106 CFU/ml和8×106 CFU/ml,对照组小鼠给予等量生理盐水;感染12h后颈动脉放血处死动物,并离心分离获得上层血清;酶联免疫吸附测定法检测感染后小鼠血清中转化生长因子β(TGFβ)、基质金属蛋白酶2/9(MMP2/9)、细胞凋亡蛋白Bax/Bcl2及炎症因子和半胱氨酸天冬氨酸蛋白酶Caspase3/9蛋白的表达变化。结果感染后小鼠血清中炎症因子白细胞介素IL-1、IL-2、IL-4、IL-10及TGFβ、基质金属蛋白酶家族MMP2/9表达明显升高,与对照组相比,差异有统计学意义(P<0.01),抑制凋亡蛋白Bcl2水平明显降低,差异有统计学意义(P<0.01),而促凋亡蛋白Bax、Caspase3/9水平明显升高,差异有统计学意义(P<0.01),且重度感染组变化明显强于轻度感染组,差异有统计学意义(P<0.01)。结论猪链球菌2型感染后小鼠存在明显的炎症反应和细胞凋亡现象,且具有剂量依从性作用。
Objective To study the effect of Streptococcus suis type 2 (SS2) infection on the expression of TGFβprotein, and to provide reference for vaccine development and drug rescue. Methods Thirty clean Kunming mice were randomly divided into control group, mild infection group and severe infection group. Mice in mild and severe infection groups were injected intraperitoneally with 2 × 106 CFU / ml streptococcus suis type 2 8 × 106 CFU / ml, the control group mice were given the same amount of saline; animals were sacrificed after carotid artery bleeding 12h after infection and centrifuged to obtain the upper serum; ELISA was used to detect the serum levels of transforming growth factor β The expression of TGFβ, MMP2 / 9, Bax / Bcl2 and inflammatory cytokines and Caspase3 / 9 protein were detected. Results The levels of inflammatory cytokines IL-1, IL-2, IL-4, IL-10, TGFβ and MMP2 / 9 in the serum of infected mice were significantly higher than those of the control group (P <0.01). The level of Bcl-2 protein was significantly decreased (P <0.01), while the levels of Bax and Caspase3 / 9 were significantly increased (P <0.01) P <0.01), and the changes in severe infection group were significantly stronger than those in mild infection group, the difference was statistically significant (P <0.01). Conclusion The mice infected with Streptococcus suis type 2 have obvious inflammatory reaction and apoptosis phenomenon, and have dose-dependent effect.
