目的:探讨姜黄素对Hela细胞(人宫颈癌细胞)增殖周期各时相的影响,为宫颈癌化疗寻求有效药物。方法:应用MTT比色法和流式细胞术测定不同浓度姜黄素(0、10、20和40 mg.L-1)对Hela细胞增殖的抑制率和细胞周期各时相的变化,并应用电子显微镜观察Hela细胞的亚细胞改变。结果:0、10、20和40 mg.L-1姜黄素对Hela细胞增殖的抑制率分别为3.0%、21.4%、32.8%和49.2%,且呈剂量依赖性;流式细胞术显示,G1期细胞增多,S期细胞减少,G2/M期细胞相对增多;细胞结构显示,细胞空化、染色质凝集、凋亡小体增多。结论:姜黄素能够抑制HeLa细胞增殖,阻止G1期细胞向S期转化的进程,促进Hela细胞凋亡。
Objective: To investigate the effect of curcumin on the phases of the proliferation cycle of Hela cells (human cervical cancer cells) and to find an effective drug for cervical cancer chemotherapy. Methods: MTT assay and flow cytometry were used to determine the inhibitory effects of different concentrations of curcumin (0, 10, 20 and 40 mg.L-1) on the proliferation of Hela cells and the changes of cell cycle phases. Subcellular changes of Hela cells were observed with a microscope. Results: The inhibitory rates of 0, 10, 20 and 40 mg.L-1 curcumin on Hela cell proliferation were 3.0%, 21.4%, 32.8% and 49.2%, respectively, in a dose-dependent manner. Flow cytometry Increased senescence, S phase cells decreased, G2 / M phase cells relative increase; cell structure, cell cavitation, chromatin condensation, apoptotic bodies increased. CONCLUSION: Curcumin can inhibit the proliferation of HeLa cells and prevent the progression of G1 phase cells to S phase and promote the apoptosis of Hela cells.
