目的观察鞘脂酶激活蛋白(sphingolipid activatorproteins,Saposins)在年龄相关性白内障晶状体上皮细胞的表达。方法收集20例年龄相关性白内障和10例正常透明晶状体的前囊膜(中央5.5~6.0mm直径区域),采用免疫组织化学染色法检测晶状体上皮细胞中鞘脂酶激活蛋白的表达并计算灰度值。结果两组晶状体上皮细胞中均有鞘脂酶激活蛋白的表达。与正常透明晶状体组相比,Saposin A(145.03±11.96)、Saposin C(139.78±7.99)和Saposin D(144.30±8.22)在年龄相关性白内障晶状体上皮细胞的表达量增加(P均<0.01),SaposinB(132.59±16.91)的表达量无明显增加(P>0.05)。结论鞘脂酶激活蛋白可能在晶状体老化和年龄相关性白内障形成过程中起重要作用。
Objective To observe the expression of sphingolipid activator proteins (Saposins) in age-related cataractous lens epithelial cells. Methods 20 cases of age-related cataract and 10 cases of normal transparent lens anterior capsule (central 5.5 ~ 6.0mm diameter area) were collected. The expression of sphingosinephric protein in lens epithelial cells was detected by immunohistochemical staining and the gray scale value. Results Both groups of lens epithelial cells had the expression of sphingosine-activated protein. The expression of Saposin A (145.03 ± 11.96), Saposin C (139.78 ± 7.99) and Saposin D (144.30 ± 8.22) in the age-related cataractous lens epithelial cells increased compared with that of the normal transparent lens group (all P <0.01) The expression of SaposinB (132.59 ± 16.91) had no significant increase (P> 0.05). Conclusion Sphingosine kinase may play an important role in the process of lens aging and age-related cataract formation.
