荣肝合剂对刀豆蛋白A诱导急性免疫性肝损伤小鼠的保护作用

目的探讨荣肝合剂对刀豆蛋白A(ConA)介导的急性免疫性肝损伤小鼠的保护作用及其机制。方法乙型肝炎病毒转基因小鼠60只,随机分为模型组、空白组、荣肝合剂组、茵陈蒿汤组、茵陈组、联苯双酯组,每组10只,采用ConA尾静脉注射法制作急性免疫性肝损伤模型。造模前14天,模型组、空白组给予生理盐水灌胃,其余组每日分别给予荣肝合剂、茵陈蒿汤、单味茵陈煎液、联苯双酯溶液灌胃。末次灌胃给药后1h,空白组给予磷酸盐缓冲液尾静脉注射,其余各组按照3μg/g体重尾静脉注射ConA造模。造模给药后8h处死动物取血或组织标本检测丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、总胆红素(TBiL)、乙型肝炎病毒载量(HBVDNA)、乙型肝炎表面抗原(HBsAg)、超氧化物歧化酶(SOD)、丙二醛(MDA)等指标。结果与模型组及其他给药组比较,荣肝合剂可降低小鼠ALT、AST、TBiL水平(P<0.05或P<0.01);与联苯双酯组、茵陈组、茵陈蒿汤组相比较,荣肝合剂可降低小鼠HBVDNA载量,降低肝组织MDA含量,提高肝组织的SOD水平,其余各组均无此作用。结论荣肝合剂的降酶作用优于单味中药茵陈、茵陈蒿汤及联苯双酯,其作用机制是通过抑制HBVDNA,减轻肝细胞膜脂质过氧化反应的损害,起到肝保护作用。 Objective To investigate the protective effect and mechanism of Ronggan mixture on ConA-induced acute immunological liver injury in mice. Methods Sixty transgenic mice with hepatitis B virus were randomly divided into model group, blank group, Ronggan Mixture group, Yinchenhao Decoction group, Yinchen group and Bifendate group, with 10 mice in each group. ConA tail vein Injection of acute immune liver injury model. 14 days before modeling, the model group and the blank group were given normal saline gavage, the rest were given Ronggan mixture, Yinchenhao decoction, single Yincao decoction and bifendate solution respectively. One hour after the last intragastric administration, the blank group was given phosphate buffered saline (TAX), and the rest were injected with ConA according to the tail vein of 3μg / g body weight. The animals were sacrificed at 8h after model administration to determine the levels of ALT, AST, TBiL and HBV ( HBVDNA), hepatitis B surface antigen (HBsAg), superoxide dismutase (SOD), malondialdehyde (MDA) and other indicators. Results Compared with model group and other administration groups, Ronggan Mixture can reduce the levels of ALT, AST and TBiL in mice (P <0.05 or P <0.01); Compared with Bifendate group, Yinchen group, Yinchenhao Decoction group In contrast, Ronggan mixture can reduce the HBVDNA load in mice, reduce the content of MDA in liver tissue and increase the level of SOD in liver tissue, but no effect in other groups. Conclusion The effect of reducing the enzyme of Ronggan Mixture is better than that of single Chinese herbal medicine Yinchen, Yinchenhao Decoction and Bifendate. Its mechanism is that it can play a protective role on the liver by inhibiting HBVDNA, reducing the damage of lipid peroxidation of liver cell membrane .
