研究纤维素酶辅助提取茵陈总黄酮的工艺优化条件,用紫外-可见比色法和芦丁为标准品测定茵陈总黄酮的得率。通过单因素实验和正交实验研究酶用量、酶解温度、酶解时间对黄酮得率的影响及工艺优化条件。研究结果表明影响茵陈总黄酮得率的因素主次顺序为:酶解温度>酶用量>酶解时间,纤维素酶辅助提取茵陈总黄酮的最优提取条件为:纤维素酶用量为0.15mg/mL,酶解过程的温度为50℃,酶解过程的时间为1.5h,此时黄酮的得率为4.21%。
The optimum conditions for cellulase-assisted extraction of total flavonoids from capillaris were studied. The yield of total flavonoids from capillaris was determined by UV-Vis and rutin as standards. The effects of enzyme dosage, enzymatic temperature and enzymolysis time on flavonoid yield and process optimization conditions were studied by single factor experiment and orthogonal experiment. The results showed that the order of the factors affecting the yield of total flavonoids of capillaris was enzymatic hydrolysis temperature> enzyme dosage> enzymolysis time, and the optimal extraction conditions for the extraction of total flavonoids from capillaris were: the dosage of cellulase was 0.15 mg / mL, the temperature of enzymolysis is 50 ℃, the time of enzymolysis is 1.5h, the yield of flavonoids is 4.21%.
