采用Franz 扩散池及离体大鼠皮肤研究了桉叶油与丙二醇对5 氟脲嘧啶的渗透促进作用。丙二醇几无促渗作用,桉叶油使药物透过量增加60 倍( Kp = 62 .80 ±10 .0 cm/h ×10 - 3) 。将桉叶油与1 % 丙二醇配合使用时具轻微的协同作用(P< 0 .05) ,药物渗透性增加至68 倍( Kp = 72 .2 ±2 .32 cm/h ×10 - 3) ,但随丙二醇用量增加至50 % ,促渗效果明显减小( Kp = 19 .80 ±0 .65 cm/h ×10 - 3 , P> 0 .05) 。在含有3 % ~10 % 桉叶油及10 % 丙二醇的1 % 5氟脲嘧啶羟丙甲纤维素凝胶中,药物经皮渗透速度进一步降低,随桉叶油用量增加促渗作用略有增强,当两种促进剂用量相等(10 % ) 时,与仅含10 % 丙二醇凝胶剂比较,渗透量仅增加5 .4 倍,Kp = 16 .40 ±0 .88 cm/h ×103 ,表明促进剂应用的浓度依赖性以及制剂因素的影响。
Franz diffusion cells and isolated rat skin were used to study the effects of eucalyptus oil and propylene glycol on permeation of 5-fluorouracil.
